Bisphosphonate inhibitors of squalene synthase protect cells against cholesterol‐dependent cytolysins

چکیده

Certain species of pathogenic bacteria damage tissues by secreting cholesterol-dependent cytolysins, which form pores in the plasma membranes animal cells. However, reducing cholesterol protects cells against these cytolysins. As first committed step biosynthesis is catalyzed squalene synthase, we explored whether inhibiting this enzyme protected We synthesized 22 different nitrogen-containing bisphosphonate molecules that were designed to inhibit synthase. Squalene synthase inhibition was quantified using a cell-free assay, and validated computer modeling binding The bisphosphonates then screened for their ability protect HeLa caused cytolysin, pyolysin. most effective reduced pyolysin-induced leakage lactate dehydrogenase into cell supernatants >80%, cytolysis from >75% <25%. A novel bisphosphonate inhibitor of squalene synthase combined with a statin or a nitrogenous bisphosphonate in vitro.

Statins and nitrogenous bisphosphonates (NBP) inhibit 3-hydroxy-3-methylglutaryl-coenzyme-A reductase (HMGCR) and farnesyl diphosphate synthase (FDPS), respectively, leading to depletion of farnesyl diphosphate (FPP) and disruption of protein prenylation. Squalene synthase (SQS) utilizes FPP in the first committed step from the mevalonate pathway toward cholesterol biosynthesis. Herein, we have...

Squalene synthase-deficient mutant of Chinese hamster ovary cells.

Squalene synthase (farnesyldiphosphate:farnesyldiphosphate farnesyltransferase, EC 2.5.1.21) converts farnesyl pyrophosphate to squalene, the first metabolic step committed solely to the biosynthesis of sterols. Using a fluorescence-activated cell sorting technique designed to screen for cells defective in the regulated degradation of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase, w...

The squalestatins, novel inhibitors of squalene synthase produced by a species of Phoma. III. Biosynthesis.

The biosynthetic origin of the carbon and oxygen atoms of the novel fungal secondary metabolite 1 was studied. Incorporation studies with single and multiple labelled 13C precursors indicated that the major portion of the molecule was derived from two polyketide chains made up of acetate units. One of the chains had benzoic acid (which can be derived from phenylalanine) as a starter unit. The r...

Biphenylquinuclidines as inhibitors of squalene synthase and growth of parasitic protozoa.

In this paper we describe the preparation of some biphenylquinuclidine derivatives and their evaluation as inhibitors of squalene synthase in order to explore their potential in the treatment of the parasitic diseases leishmaniasis and Chagas disease. The compounds were screened against recombinant Leishmania major squalene synthase and against Leishmania mexicana promastigotes, Leishmania dono...

Glucosylceramide synthesis and synthase expression protect against ceramide-induced stress.

Ceramides (Cers), critical for epidermal barrier function, also can inhibit keratinocyte proliferation, while glucosylceramides (GlcCers) exert pro-mitogenic effects. Since alterations in Cer-to-GlcCer ratios appear to modulate cellular growth versus apoptosis, we assessed whether keratinocytes up-regulate GlcCer synthesis as a protective mechanism against Cer-induced stress. Exogenous sphingom...
